For a short, standard tuk tuk trip where you negotiated the fare beforehand or used the meter, a tip is not strictly required. However, if the fare is 300 Rupees, simply handing the driver a 500 Rupee note and telling them to keep the change, or explicitly giving a 50 to 100 Rupee tip, is a wonderful gesture. Given the current economic situation, that tiny extra amount means very little to a tourist but is massively appreciated by the hardworking local drivers.
