Yes, there are a few private AC luxury mini buses and standard government buses that run late into the night between Colombo and Kandy. You can usually catch them at the Bastian Mawatha terminal.

While they are generally safe, the driving can be incredibly fast and reckless on the empty, winding mountain roads at night. If you want a comfortable, stress free journey, taking the train during the day is vastly superior.
