Are there public buses running at 5 AM from Galle that will get us to Mirissa Harbor in time for the 6:30 AM whale watching tours.

The Route 32 buses run 24/7, so technically yes, you can catch one at 5 AM from Galle central bus stand. However, the early morning schedules are very erratic. You risk missing your expensive boat tour if the bus is delayed. It is much safer to pre-arrange a tuk-tuk the night before to guarantee you arrive by 6:00 AM.

Do not risk it. The boats leave exactly at 6:30 AM and do not wait for anyone.
