The most direct and comfortable way from Colombo airport to Kandy is booking a private taxi. The drive takes about 3 hours depending on traffic and costs roughly 45 to 60 USD. You can arrange this via your hotel in advance or use the airport taxi counters in the arrivals hall. If you are on a strict budget, you can take a taxi to the Kadawatha bus station near Colombo, and from there catch a luxury air conditioned intercity bus straight to Kandy for just a few dollars.
If you arrive in the early morning, you could also take an Uber from the airport to the Colombo Fort Railway Station, and catch the morning scenic train up to Kandy. It takes a bit longer, but the views as you climb into the hills are incredible.
Do not take a standard tuk tuk all the way to Kandy. It is a long, dusty ride up the mountains and is not safe for highway driving over that distance. Stick to an enclosed car or the express buses.
